General description

The MISSION TRC3 ORF GFP Control Vector is a lentivirus plasmid vector. The vector contains an ORF insert expressing TurboGFP, and is useful as a positive (or negative control) in experiments using the MISSION TRC3 LentiORF Library Collection in arrayed or pooled format, including our latest Whole Genome LentiORF Pool. TurboGFP is an improved variant of the green fluorescent protein copGFP cloned from copepoda Pontellina plumata.

This positive control can be used for measuring transduction efficiency and optimizing ORF delivery in cell lines that do not survive any puromycin exposure. The GFP Control contains a gene encoding TurboGFP driven by the EF1a promoter. This control provides fast visual confirmation of successful transduction. Ampicillin and puromycin antibiotic resistance genes provide selection in bacterial or mammalian cells respectively.

In addition, self-inactivating replication incompetent viral particles can be produced in packaging cells (HEK293T) by co-transfection with compatible packaging plasmids. The MISSION TRC3 ORF GFP Control Vector is provided as 10 μg of plasmid DNA in Tris-EDTA (TE) buffer at a concentration of 500 ng/μl.
