Application

Casein Blocking Buffer 10x has been used in:

  • western blotting[1][2][3]
  • enzyme linked immunosorbent assay (ELISA)[4]
  • immunohistochemistry[5]

Features and Benefits

  • Better blocking performance than dried milk or BSA
  • Easy-to-use concentrate
  • Compatible with neutral and positively-charged membranes
  • Compatible with chemiluminescent and colorimetric detection assays

General description

Casein is a phosphoprotein found in milk. It is classified into four subtypes, including αS1-, αS2-, β-, and κ-caseins.[6]
Sigma’s 10x Blocking Buffer is a casein-based blocking agent routinely used to reduce non-specific binding of detection reagents[6] to the Northern and Southern Blot membranes.

Preparation Note

Dilute 10x Blocking Buffer with molecular biology grade water before use.

Questions

1–7 of 7 Questions
  1. Hi, how much time can be the casein blocking buffer 10X stored from its opening?

    1 answer
    1. This product does not have an expiration date once opened. The datasheet indicates the following regarding storage and stability:

Storage/Stability: Store the 10X Casein Blocking Buffer at room temperature. It is also stable when stored at 2–8°C. Casein may precipitate over time, which is normal and should not impact the performance of the 10X Blocking Buffer. If precipitation is observed, resuspension of the solids can be achieved by inverting the bottle several times or by vortexing for a few seconds.

  2. Hi, would it be possible to know the % of casein in your Casein Blocking Buffer 10X Cat# B6429? Thank you.

    1 answer
    1. The composition of this product is proprietary and cannot be disclosed. However, it can be stated that the product is a blocking reagent derived from bovine milk casein and does not contain any serum albumin.

  3. whats the percentage casein in 10X of B6429-500ml ?

    1 answer
    1. Unfortunately, the concentration of casein is considered proprietary.

  4. Will my RNA samples and probes be safe with this buffer if we are using horseradish peroxidase and chemiluminescent detection?

    1 answer
    1. The 10X Casein Blocking Buffer is recommended for both colorimetric and chemiluminescent detection. It is used as a blocking solution for detection, which occurs after hybridization and is suitable for Northern and Southern applications.

      Helpful?

  5. What is the pH? What is the counter ion? Is there saline salt in the preparation (0.15M)?

    1 answer
    1. Unfortunately, the composition of this product is proprietary and can't be disclosed.

      Helpful?

  6. What is the % w/v casein in this solution?

    1 answer
    1. The casein concentration is 1.8% of the total solution.

      Helpful?

  7. What is the solution solvent? What grade of reagent is this in terms of sterility and associated testing?

    1 answer
    1. As mentioned in the product detail page, this product is a molecular biology-grade product. A solution is prepared by diluting the 10x Blocking Buffer with molecular biology-grade water.
