Nectin-2 (UniProt: Q92692; also known as Herpes virus entry mediator B, Herpesvirus entry mediator B, HveB, Nectin cell adhesion molecule 2, Poliovirus receptor-related protein 2, CD112) is encoded by the NECTIN2 (also known as HVEB, PRR2, PVRL2) gene (Gene ID: 5819) in human. Nectin-2 is a single-pass type I membrane protein that displays ubiquitous expression. It is a Ca2+-independent cell-cell adhesion molecule that regulates cell adhesion between epithelial cells through the formation of transdimers between adjacent cells. Nectin-2-mediated cell adhesion is shown to induce the formation of E-cadherin-based adherens junctions followed by the formation of claudin based tight junctions. Nectin-2 also serves as a modulator of T-cell signaling and it can be either a co-stimulator of T-cell function, or a co-inhibitor depending on the receptor it binds to. Upon binding to CD226, it stimulates T-cell proliferation and cytokine production, including that of IL-2, IL-5, IL10, IL-13, and Interferon-g. Its interaction with PVRIG, inhibits T-cell proliferation. These interactions are shown to be competitive in nature. Over-expression of Nectin-2 has been reported in breast and ovarian cancer tissues and in several cancer cell lines. It is synthesized with a signal peptide (aa 1-31), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 32-360), a transmembrane domain (aa 361-381), and a cytoplasmic domain (aa 382-538). Nectin-2 has one Ig-Like V-type domain (aa 32-156) and two Ig-like C2 type domains (aa 162-256 and 261-345).
