本製品はE. coli血清型O111:B4から抽出したTCAです。ソース菌株は私的コレクションのものです。このLPS血清型はB-細胞の刺激および ヒト肝細胞におけるNOSの誘発に使用されています。

アプリケーション

リポ多糖（LPS）は、グラム陰性菌の細胞壁に特徴的な成分です。LPSとその成分である脂質Aは、一般的な病原体関連分子パターン（PAMP）を認識するトール様受容体タンパク質ファミリーの1つである、トール様受容体4（TLR4）による先天的な免疫システムの細胞を刺激します。

生物化学的／生理学的作用

リポ多糖類（LPS）は膜の外層に局在し、 非カプセル化菌株ではセル表面に露出しています。LPSは外膜の完全性に寄与し、胆汁酸塩および親油性抗生物質の作用に対して細胞を保護します。

調製ノート

この製品は水（5 mg/mL）または細胞培地（1 mg/mL）に可溶で、濁りのある薄黄色の溶液が得られます。より高濃度の溶液（20 mg/mL）は、まだ濁りがありますが、生理食塩水中でボルテックスして70～80oCで温めて得ることができます。リポ多糖類はあらゆる溶媒でミセルを形成する分子です。水およびリン酸緩衝生理食塩水では溶液に濁りが見られます。有機溶媒では透明な溶液は得られません。メタノールでは浮遊物がある不透明な懸濁液となりますが、水の場合は均一な濁りのある溶液となります。
