このインヒビターはトリプシンに対して作用し、キモトリプシンおよびプラスミンに対してこれよりも弱い作用を及ぼします。 このインヒビターはトリプシン、血漿カリクレイン、凝固因子Ｘと同様のメカニズムをもってプロテアーゼも阻害します。トリプシンインヒビターは、メタロプロテアーゼ、組織ベースのカリクレイン、酸プロテアーゼまたはチオプロテアーゼに対しては作用しません。 このインヒビターは、プロテアーゼ活性部位の1：1の化学量論合成物を生成することで作用し、次にインヒビターの単一のアルギニン-イソロイシン結合が開裂します。 阻害は可逆性でpH依存性です。
Preparation Note
カルシウムとマグネシウムを含まないダルベッコのリン酸緩衝生理食塩水で調製されています。
トリプシンインヒビターは水およびリン酸バッファーに10 mg/mLの濃度で溶けます。 また、平衡塩溶液に1 mg/mLの濃度で溶け、無血清培地にも溶けます。 濃度が10 mg/mLを超える濃縮溶液は濁ることがあり、黄色からアンバーを呈します。 細胞をトリプシン化した後、解離に使用したトリプシン溶液1 mLに対し1 mLのトリプシンインヒビター溶液に1 mg/mLで再懸濁させます。 次にこの細胞懸濁液を1000 rmpで遠心分離し、細胞ペレットを生成します。
溶液は2～8℃で短期間保存したとき活性を保持できます。溶液はアリコートを-20℃で凍結させると安定します。

I isolate keratinocytes from human epidermis. For this purpose, I use 4 ml of 5x trypsin/EDTA (Gibco 15400054). I would like to use your trypsin inhibitor T6414 for the inactivation of trypsin. Can you tell me what concentration is required for this?

I am trying to find the activity level on your website for this specific item: T6414. I tried to search on your website and Sample COA but it doesn’t seem to have one, can you provide me a ball park range for what the activity level is for the item?

This product is a 1mg/mL solution of Trypsin inhibitor. One milligram of the Trypsin Inhibitor powder used to prepare this solution will inhibit between 1.0 - 3.0 mg of trypsin. The usage instructions provided in the product datasheet recommend adding 1ml of this solution per milligram trypsin. Please see the link below to review the datasheet:
