Application

El sustrato TMB se utiliza en procedimientos ELISA en los que se utilizan conjugados de peroxidasa de rábano picante. El sustrato TMB desarrolla un producto de reacción azul soluble que puede leerse a 370 o 655 nm. La reacción se detiene con ácido, y se forma un producto de reacción amarillo que permite una medición precisa de la intensidad a 450 nm.

Cómo utilizar nuestro sustrato TMB:
  • Nuestro sustrato TMB se suministra como sustrato de peroxidasa listo para usar que contiene TMB en un tampón ligeramente ácido. Antes de la reacción con la peroxidasa, el sustrato debe ser una disolución de incolora a verde azulada clara. El sistema sustrato desarrolla un producto de reacción azul cuando reacciona con la peroxidasa.
  • Una vez alcanzado el punto final del ensayo con la adición de ácido, se mide la intensidad del sustrato TMB a 450 nm.
  • Nuestro sustrato TMB está recomendado para los procedimientos ELISA (micropocillo) y se ha utilizado en diversos ensayos como sustrato del conjugado de anticuerpos y peroxidasa de rábano picante para la detección de la glucoproteína del virus Hendra, la tetracosáctida, la inmunoglobulina A secretora (sIgA) y las trampas extracelulares de neutrófilos (NET).
  • El sistema de sustrato líquido de 3,3′,5,5′-tetrametilbencidina (TMB) no se recomienda para aplicaciones de membrana. Dado que el sustrato TMB rinde un producto de reacción soluble, no se recomienda para histoquímica ni para inmunotransferencia.
Nota: El sustrato TMB es sensible a la luz y debe protegerse de la luz solar directa o de fuentes UV.

    1. There is no set time at which the reaction must be stopped. Depending on the peroxidase activity measured, the color may form very quickly or slowly. Kindly adhere to the instructions provided in our datasheet as closely as possible.

    1. No hydrogen peroxide should be added to product T0440. The product, which contains  a peroxidase substrate,  already contains hydrogen peroxide.  This solution is ready to use for ELISA assays.
