Protein Mass Spectrometry

The Sigma-Aldrich enzyme portfolio provides a comprehensive and versatile toolkit for proteomics mass spectrometry workflows. Solutrypsin, a highly purified and soluble form of trypsin, offers consistent and efficient cleavage at the carboxyl side of lysine and arginine residues. Its superior solubility and reduced autolysis make it ideal for high-throughput and reproducible digestions, especially in complex or denaturing sample conditions. ASP-N complements this by targeting the N-terminus of aspartic acid residues, expanding sequence coverage in acidic regions often missed by trypsin alone. Glu-C adds further depth by cleaving at glutamic and aspartic acid residues, supporting detailed mapping of post-translational modifications. Lysarginase, which cleaves after lysine and arginine with a different specificity profile than trypsin, offers orthogonal cleavage patterns that enrich peptide diversity and proteome coverage. Together, these enzymes enable high-resolution, in-depth proteome analysis across a wide range of experimental conditions.

Enzyme Reagents
Reliable and efficient protein digestion with selective and pure sequencing grade proteases is a crucial step when preparing proteomics samples. We offer a wide range of both natural source and recombinant proteases including endoproteinases like Trypsin (T1426), Chymotrypsin (C6423), Lys-C (324715), Glu-C (324713), Asp-N (324708), as well as a variety of Aminopeptidases (A8200) and Carboxypeptidases (C9584). Additionally, SOLu-Trypsin is a ready-to-use recombinant trypsin solution (EMS0004). This Advanced Proteomics Grade enzyme is solution stable for mass spectrometry, designed to be stable in solution when refrigerated, and SOLu-Trypsin can be used immediately without preparation.
