General description

TransPlex, a Whole Transcriptome Amplification (WTA) method, allows for representative amplification of nanogram quantities of total RNA in less than 4 hours without 3′-bias. Microgram quantities of amplification product generated from tissue, cultured cells, formalin-fixed samples, or serum are suitable for downstream applications such as qPCR and microarray analyses.

Principle

The WTA process involves two steps. In the first step, sample RNA is reverse transcribed with non-self-complementary primers composed of a quasi-random 3′ end and a universal 5′ end. As polymerization proceeds, displaced single strands serve as new templates for primer annealing and extension. The resultant OmniPlex cDNA library, comprised of random, overlapping 100 - 1000 base fragments flanked by universal end sequence, is then amplified by PCR with the universal primer to produce WTA product.

  3. What is the concentration of the dNTPs supplied with the TransPlex® Whole Transcriptome Amplification Kit, Product WTA1?

    The dNTP Mix (D6193) is supplied as 10 mM each.

  4. Is the TransPlex® Whole Transcriptome Amplification Kit, Product WTA1, also applicable for ribosomal RNA and/or small RNA amplification?

    Using the kit as is, Ribosomal RNA does appear to amplify. However, we have not quantitated how well it is amplified.

  5. What is the difference between WTA and WGA?   I use the TransPlex® Whole Transcriptome Amplification Kit, Product WTA1.

    The starting material for WTA is RNA, whereas for WGA it is genomic DNA.  After the RNA is reverse-transcribed into the DNA complement by WTA, the OmniPlex library is formed. This library is in turn amplified by PCR.

  6. Can TransPlex® Whole Transcriptome Amplification Kit, Product WTA1, be used to amplify archival fixed tissue or degraded samples?

    Yes, the WTA kit effectively amplifies degraded RNA, including formalin-fixed and paraffin-embedded samples.

  7. Can TransPlex® Whole Transcriptome Amplification Kit, Product WTA1, be used for downstream applications?

    Upon creation of a cDNA library, quantitative qPCR or microarray analysis may be performed
