General description

Endothelial cell growth supplement (ECGS) comprises acidic fibroblast growth factor (aFGF) and basic FGF (bFGF).

Application

Endothelial cell growth supplement from bovine neural tissue has been used as a supplement in Dulbecco′s modified Eagle′s medium supplement and F-12K media supplement for human umbilical vein endothelial cells culture.

Biochem/physiol Actions

Endothelial cell growth supplement (ECGS) improves proliferation of human saphenous vein endothelial cells (HSVEC) and human umbilical vein endothelial cells (HUVEC). It coordinates with interleukin 1 (IL-1) and mediates matrix remodelling in vascular smooth muscle cells (VSMCs). It is essential for endothelial cells proliferation and adipose-derived stem cell (ASC) differentiation. An extract of bovine neural tissue containing growth promoting factors for vascular endothelial cells of mammalian origin. ECGS has also been reported to be beneficial as a media supplement for the fusion and growth of hybridoma cells in monoclonal antibody production.

Preparation Note

ECGS is an extract of bovine neural tissue containing growth promoting factors for vascular endothelial cells of mammalian origin. Endothelial cell growth supplement is prepared using a modification of the method of Maciag, et al., and is lyophilized from a sterile solution containing NaCl and streptomycin sulfate.

Analysis Note

The proliferative activity of endothelial cell growth supplement (ECGS) is tested in culture using fetal bovine heart endothelial cells (ATCC CRL 1395) seeded at low density. The EC50 is defined as the effective concentration of growth factor that elicits a 50% increase in cell growth in a cell based bioassay.
