桑格测序 早期,分析化学方法便已能够测定核酸的组成。然而,弗雷德·桑格及其同事开发了一系列方法——最初采用放射性标记的消化片段和二维分馏技术——从而获得了首批完整的核酸序列。 该领域在随后的几十年里持续取得进展,每一项改进都为不断扩大的蛋白质和基因组测序库增添了新内容。这些改进包括利用凝胶电泳结合毛细管电泳按长度分离核苷酸。此外,荧光标记核苷酸的引入以及对每个核酸片段进行的自动化计算机分析,共同构成了现代桑格测序法。 ##
