## 桑格测序
早期，分析化学方法便已能够测定核酸的组成。然而，弗雷德·桑格及其同事开发了一系列方法——最初采用放射性标记的消化片段和二维分馏技术——从而获得了首批完整的核酸序列。 该领域在随后的几十年里持续取得进展，每一项改进都为不断扩大的蛋白质和基因组测序库增添了新内容。这些改进包括利用凝胶电泳结合毛细管电泳按长度分离核苷酸。此外，荧光标记核苷酸的引入以及对每个核酸片段进行的自动化计算机分析，共同构成了现代桑格测序法。
## DNA测序方法
尽管自该技术诞生以来，DNA测序方法一直在不断改进，但现代DNA测序平台仍需依赖并使用若干基本组件。 DNA制备通常包括从宿主生物体中分离和纯化DNA。其他关键组分，包括游离DNA碱基、DNA引物、带有荧光标记的修饰DNA碱基（终止碱基）以及DNA聚合酶，会被共同加入到一个反应管中。装有所有这些组分的反应管经过一系列加热和冷却步骤后，将产生一个由相对于目标全长DNA序列而言较短的DNA片段组成的文库，每个片段末端都带有荧光末端标记的终止碱基。 这些含有荧光末端标记核苷酸的新DNA链将按长度进行分离，通过毛细管，并按大小排列。随后，利用激光激发每条链上的荧光碱基，同时摄像头捕获该信号。最后，通常由计算机将收集到的信息组装成完整的DNA序列。
