# 热原测试
热原检测确定肠外制药产品中是否存在热原，受食品药品管理局 (FDA)、美国药典 (USP) 或欧洲药典 (EP) 等组织的多项标准监管。产品无菌并不意味着不含热原。因此，声称无菌的药品也必须进行热原检测，以防止患者出现发热反应。
热原污染可能发生在药品、生物治疗剂和医疗器械的生产或使用过程中，但热原的存在也可能是产品的固有特性，例如疫苗中的佐剂或合成脂肽。

热原是一种通过激活先天性免疫系统导致人类或动物体温升高（发热反应）的物质。它们是由微生物和非微生物物质组成的一组不同的污染物。热原可分为两类：内毒素和非内毒素热原（NEPs）。
内毒素是存在于革兰氏阴性细菌中的物质。非内毒素致热原是其他微生物物质，包括来自革兰氏阳性细菌或病毒的致热原以及来自酵母和真菌的致热原。非微生物致热原物质也可能来自橡胶颗粒、微小塑料颗粒或弹性体中的金属化合物。
目前有多种检测致热原的方法。这些方法可根据其检测的污染物类型以及进行检测时对动物材料的需求进行分类，如下表所示：

兔子热原测试（RPT）包括测量兔子静脉注射受测产品后的体温升高情况。RPT 可给出定性结果，灵敏度很低。由于兔子在反复注射后会对热原产生耐受性，或在进行试验时兔子会产生应激反应，因此该试验的稳健性也很有限。

[单核细胞活化检验（MAT）](https://www.sigmaaldrich.com/SG/zh/products/industrial-microbiology/pyrogen-testing)是检测内毒素和非内毒素热原的动物方法的替代方法。单核细胞活化检验通过将单核细胞与被测样品一起孵育来模拟人体的免疫反应。如果存在热原，单核细胞就会被激活并产生炎症分子--细胞因子，导致发热反应。细胞因子随后通过免疫测定（ELISA）进行检测，该方法涉及特异性抗体和酶促颜色反应。
