固相萃取（SPE）是一种专为色谱分析（如HPLC、GC、TLC）前的快速、选择性样品制备和纯化而设计的技术。在SPE过程中，通过萃取、分配和/或吸附到固相固定相上，从液态样品中分离出一种或多种分析物。
通过SPE进行样品制备，可将样品的原始基质转变为更简单的基质环境。这使得样品更适合后续的分析色谱，通常能简化并提高最终的定性与定量分析效果。更简单的样品基质还能降低对分析系统的要求，从而可能延长系统的使用寿命。
优化后的 SPE 流程可帮助您：
- 转换样品基质，使其与目标色谱方法更兼容。
- 浓缩分析物（痕量富集），以提高灵敏度。
- 去除在色谱分析过程中导致高背景、误导性峰和/或灵敏度较低的干扰物。
- 保护分析柱免受污染物影响。
- 实现萃取过程的自动化。
## SPE 是如何工作的？
在 SPE 中，固定相（吸附剂或树脂）通过强而可逆的相互作用与分析物或杂质结合，从而从复杂样品中可靠、快速地萃取目标分析物。
SPE 具有选择性和多功能性，因为针对不同的分析物和基质，可选用多种不同的吸附剂和洗脱条件。常见的 SPE 吸附剂包括：
- 以二氧化硅为基础的
- 反相（C18、C8、氰基、苯基）
- 正相（硅胶、二醇、NH₂）
- 离子交换（SAX、WCX、SCX）
- 碳基
- 聚合物基（各种组成、不同官能团）
