General description

MacConkey broth is a selective culture medium containing lactose. Upon degradation, lactose forms acid and gas, indicating the presence of E. coli. The gas formed is collected in Durham tubes. A change in the color of the pH indicator bromocresol purple to yellow indicates acid production. Ox bile promotes the growth of several species of intestinal bacteria and inhibits the growth of other microorganisms that do not inhabit the intestine.

Application

MacConkey broth is suitable for presumptive tests for Escherichia coli and coliform bacteria and for determining the E. coli or coliform titer of milk, water, and other materials. This medium complies with the specifications given by the harmonized methods of EP, USP, JP for Microbial Examination of Non-sterile Products: Tests for Specified Microorganisms.
