General description

A competitive enzyme immunoassay (EIA) is employed for the quantitative determination of thromboxane B2 in cell culture supernatants, saliva, urine, serum, and plasma. Recognizes human, but does crossreact with mouse, rat, rabbit, and other mammalian TXB2. In the assay, the TXB2 protein in the sample competes with a fixed amount of TXB2 conjugated to alkaline phosphatase for the limited number of binding sites on the antibody. Bound enzyme reacts with the pNpp substrate and the reaction produces yellow color, whose intensity, read at 405 nm, is inversely proportional to the concentration of TXB2 in the standards or samples. The measured optical density is used to calculate the concentration.

Biochem/physiol Actions

TXA2 is involved in platelet aggregation, vasoconstriction and reproductive functions. However, due to its short half-life of 30 seconds at 37 °C in vivo, TXA2 is hydrolyzed rapidly to TXB2. Thus, TXA2 formation is assayed by quantifying TXB2, which is more stable.

Features and Benefits

The assay is sensitive (detects less than 10.54 pg/mL), fast (total time 3 hours) and user-friendly (precoated plates eliminate the coating step, all incubations proceed at room temperature, any commercially available microwell plate reader may be used and reagents are color coded for the ease of use). The sample recovery ranges from 98% (human saliva) to 113% (human serum) and there is negligible cross-reactivity with 20-25 related eicosanoid compounds tested (from <0.1 to 2%).
