General description

Escort is a (1:1) liposome formulation of the cationic lipid DOTAP and the neutral lipid DOPE . This liposome-forming compound is used for transfection of nucleic acids into eukaryotic cells.


Suitable for transient and stable transfection of nucleic acids into cultured eukaryotic cells. Use approximately 15 μl Escort and 5 μg DNA per 6 cm cell culture plate. Protocol optimization provides very efficient transfection. The following cells have been successfully transfected using Escort:

Bone marrow stromal cells (human)
D. mel
Oral Squamous Cell Carcinoma (human)

Escort formulation:
1 mg/mL DOTAP
1 mg/mL DOPE
20 mM MES
150 mM NaCl (pH 6.2)


Do not freeze.


A stable complex is formed when DOTAP/DOPE is mixed with DNA in the absence of serum. The complexes are stable and can be directly added to the cell culture medium, where they fuse with the cell membrane, releasing the DNA into the cytoplasm. Note: complex formation is inhibited by serum, but once stable complexes have formed, the presence of serum is without consequence.

Other Notes

The given protocol was developed for transient transfection of HeLa cells using pSV40-CAT plasmid as a reporter gene.
