General description

The widely used MMP substrate FS-1 (Cat. No. 03-32-5032) is modified by an N-terminal lysine elongation to produce a broad-spectrum MMP and TACE substrate that displays improved water solubility and substrate sensitivities. Exhibits 2- to 9-fold higher specificity constant (kcat/Km) towards MMP-1, -8, -13, -14 when compared with FS-1 and ~2-fold higher kcat/Km towards TACE when compared with TACE Substrate II and III (Cat. Nos. 616402 and 616403, respectively). It has been used successfully in detecting T98G glioma cell surface-associated MMP activity and is shown to be resistant to proteolysis by human leukocyte elastase, trypsin, and ADAM-10.

Exhibits a 2- to 9-fold higher specificity constant (kcat/Km) towards MMP-1, -8, -13, -14 (Km (μM)= 27.5, 5.2, and 7.9, respectively) when compared with FS-1 and ~ 2-fold higher kcat/Km (778, 400 M-1 s-1) towards TACE when compared with TACE Substrate II and III (Cat. Nos. 616402 and 616403, respectively). It has been used successfully in detecting T98G glioma cell surface-associated MMP activity and is shown to be resistant to proteolysis by human leukocyte elastase, trypsin, and ADAM-10.
