General description

Recombinant, human PPI2 expressed in E. coli. Inhibitor-2 is the heat-stable, regulatory subunit of the cytosolic type 1 Ser/Thr protein phosphatase-1 (PP1). Inhibits the activity of the free catalytic subunit of PP1 (IC50 = 1 nM). Inhibition is reported to involve a "masking region" on the inhibitor that lies in close proximity to the active site of PP1 and blocks the access to substrates. Interaction of inhibitor-2 with the catalytic subunit of PP1 leads first to a rapid inhibition of the enzyme activity followed by the formation of a stable inactive complex. The inactive complex can be reactivated through phosphorylation of inhibitor-2 at Thr72 by glycogen synthase kinase-3 (GSK-3).
