Pellet Paint NF Co-Precipitant is anonfluorescent dye-labeled carrier compatiblewith fluorescent sequencing. It facilitatesrapid removal of BigDye Terminators duringthe alcohol precipitation of cycle sequencingreaction products. Cycle sequencing reactionscan be precipitated rapidly with 1 µl of carrierper reaction and centrifugation times of 10minutes. The easily visualized carrier providesa simple confirmation that precipitationhas occurred. Sequencing reaction productsare efficiently pelleted and dye-labeledterminators remain in the supernatant duringalcohol precipitation using the standardApplied Biosystems precipitation protocols.Resuspension of pelleted sequencing reactionproducts in deionized formamide can beconfirmed by checking for dissolution ofthe carrier. Pellet Paint NF Co-Precipitantis fully compatible with the ABI PRISM®BigDye Terminator Cycle Sequencing ReadyReaction. To avoid extra sample handling,Pellet Paint NF Co-Precipitant can be addeddirectly to the reaction mix, template DNA,crude PCR samples, or dilution buffer beforethe cycle sequencing reaction. Although PelletPaint NF absorbs in the UV range, accuratespectrophotometric measurements of DNA orRNA samples are possible; the absorbance ratio(provided with each package of Pellet PaintNF) can be used as a correction factor whendetermining nucleic acid concentration. PelletPaint NF Co-Precipitant has no detectableeffect on the sequencing reaction or sequenceaccuracy. Pellet Paint NF Co-Precipitant isa useful substitute for the original PelletPaint Co-Precipitant in applications wherefluorescent detection is used.
