General description

Prolow-density lipoprotein receptor-related protein 1 (UniProt Q07954; also known as A2MR, Alpha-2-macroglobulin receptor, APOER, Apolipoprotein E receptor, CD91, LRP-1) is encoded by the LRP1 (also known as A2MR, APR) gene (Gene ID 4035) in human. LRP1 is an LDLR family member expressed by vascular endothelial cells forming the blood-brain barrier (BBB), vascular mural cells (pericytes and vascular smooth muscle cells (VSMCs)), neurons, and astrocytes. LRP1 recognizes and mediates the endocytosis of more than 40 different ligands, including apolipoprotein E (apoE), APP and Aβ. LRP1 is the primary receptor mediating transport of Aβ across the BBB into circulation, thereby clearing it from the brain. LRP1 is synthesized with a signal peptide sequence (a.a. 1-19), posttranslational cleavage yields a soluble form (sLRP1) and a cell surface-bound form composed of an 515 kDa extracellular α-chain (LRP-515) and a 85 kDa single-transmembrane (a.a. 4420-4444) β-chain (LRP-85) that contains the 100-amino intracellular domain (LRPICD; a.a. 4445-4544). BACE-1, ADAM-10, ADAM-12, ADAM-17, MT1-MMP, and t-PA are shown to be involved in the shedding of LRP1 to produce sLRP1.

Specificity

Recognizes the Pro- and mature LRP-1 (a.a. 1-4544 and 20-4544, respectively), the beta-secretase-cleaved, membrane-bound LRP-85, and the gamma-secretase-cleaved intracellular domain LRPICD fragment, but not the beta-secretase-cleaved extracellular LRP-515 fragment.

Immunogen

Epitope: LRPICD.
GST-tagged recombinant human LRP1 intracellular domain (LRPICD).

Application

Anti-LRP1 Antibody, Intracellular domain is an antibody against LRP1 for use in Western Blotting.

Western Blotting Analysis: A representative lot detected the LRP-85 target band in NIH/3T3 cell lysate, as well as in mouse brain and liver homogenates (Courtesy of Professor Nicola Zambrano, University of Naples Federico II, Italy).
Western Blotting Analysis: A representative lot detected a decreased LRP-85 target band in lysates from LRP1 shRNA-treated NIH/3T3 cells (Courtesy of Professor Nicola Zambrano, University of Naples Federico II, Italy).

Quality

Evaluated by Western Blotting in NIH/3T3 cell lysate.

Western Blotting Analysis: A 1:5,000 dilution of this antibody detected the LRP-85 target band in 10 µg of NIH/3T3 cell lysate.ysate.

Target description

~80 kDa observed. Uncharacterized band(s) may appear in some lysates.

Physical form

Rabbit polyclonal antibody serum with 0.05% sodium azide.
Unpurified

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
