General description

Glucagon-like peptide-1 (GLP-1) is a 30 amino acid gut hormone secreted from the intestinal L-cells. The GLP-1 sequence is highly conserved among mammals and is released in its active form (7-36) in response to food intake. It is quickly degraded into its inactive form (9-36) after cleavage by DPPIV. GLP-1 reduces glucose levels by regulating pancreatic secretion, slowing gastric emptying and lowering the desire for food intake. Therefore, it is an important biomarker for the study of diabetes and obesity. This kit measures both the active (7-36) and inactive (9-36) forms of GLP-1.

Application

Research Category
Metabolism
This is a 3.5-hour assay at room temperature requiring 50 uL of serum, plasma or culture samples per well.
Used to detect/quantify: Glucagon Like Peptide-1

Biochem/physiol Actions

Cross Reactivty
GLP-1 is conserved in almost all mammals.
The antibody pair used in this assay is specific to GLP-1 (7-36) and (9-36) and has no cross-reactivity with GLP-2, GIP, Glucagon or Oxyntomodulin.

The short-form glucose-dependent insulinotropic polypeptide (GIP) (1-30) is released from islet alpha cells and promotes insulin secretion in a paracrine manner in vitro. However, it is not well elucidated how GIP (1-30) is involved in glucose metabolism in vivo, since

The enteroinsular axis (EIA) comprises intestinal factors (incretins) that stimulate insulin release after PO ingestion of nutrients. Glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) are the main incretins. The EIA has not been investigated in healthy neonatal foals but

Questions

1–3 of 3 Questions
  1. What is the cross reactivity to other forms of GLP-1?

    1 answer
    1. The assay detects GLP-1 (1-37, 1-36, 7-37), with a moderate affinity and exhibits 12-15% cross-reactivity.

      Helpful?

  2. Are there any instances of cross-reactivity to Liraglutide when testing patient samples treated with Liraglude in EZGLP1T-39K, considering that these patient samples read higher than control samples?

    1 answer
    1. The kit has not been tested for cross-reactivity to liraglutide. Since liraglutide is similar to GLP-1 7-37 and our kit has 15% cross-reactivity to GLP-1 7-37, there is a possibility of some cross-reactivity.

      Helpful?

  3. What information is available for the antibodies used in the GLP-1 Total ELISA kit (EZGLP1T-36K)?

    1 answer
    1. The sequence and epitope are proprietary. The capture involves a polyclonal rabbit anti GLP-1 total, while the detection uses a monoclonal mouse anti GLP-1 (7-36), and the immunogen is mid molecule.
