General description

Inflammation and immune cell response are key to maintaining homeostasis. The more we understand mechanisms of disease, the more we discover the role that inflammation plays in the etiology of disease: certainly immunological diseases, such as allergic reactions, asthma, infectious disease and an ever-growing number of conditions under the autoimmune umbrella, but also neurodegenerative disease, metabolic and cardiovascular disease and even cancer. The ability to measure multiple inflammation biomarkers simultaneously using MILLIPLEX® MAP panels, based on the Luminex xMAP technology, to establish disease profiles that can be used for screening, diagnosis and response to treatment is critical for understanding the pathogenesis of these diseases. The MILLIPLEX® MAP Cytokine/Chemokine Magnetic Bead Panel IV Premix is a 20-plex premixed kit to be used for the simultaneous quantification of all of the following analytes in serum, plasma or culture supernatant samples: BAFF/Blys, BRAK/CSCL14, CCL28, CXCL16, HMGB1, IFNβ, IL-14, IL-19, IL-24, IL-28B, IL-32α, IL-34, IL-35, IL-36β, IL-37, IL-38, MIP-4/PARC/CCL18, MPIF-1/CL23 and YKL40/CHI3L1.MILLIPLEX® MAP offers the broadest selection of analytes across a wide range of disease states and species. Once the analytes of interest have been identified, you can rely on the quality that we build into each kit to produce results you can trust. In addition to the assay characteristics listed in the protocol, other performance criteria evaluated during the validation process include: cross-reactivity, dilution linearity, kit stability, and sample behavior (e.g. detectability and stability). Specificity

Cross-reactivity between the antibodies and any of the other analytes in this panel is non-detectable or negligible.

Application

The Human Inflammation / Immunology Bead-Based Multiplex Cytokine/Chemokine Premix Assay kit IV, using the Luminex® xMAP technology, enables the simultaneous analysis of 20 cytokine and chemokine biomarkers in human serum, plasma and cell culture samples.This is an overnight or two-hour incubation assay; however, for best results an overnight incubation is recommended.A maximum of 25 μL per well of 1:4 diluted serum or plasma can be used. Tissue culture supernatant may require a dilution with an appropriate control medium prior to assay.
