General description

“Cytokine” is a general term used for a diverse group of soluble proteins and peptides which act as regulators under both normal and pathological conditions to modulate the functional activities of individual cells and tissues. These proteins also mediate direct interactions between cells and regulate processes taking place in the extracellular environment. The cytokine group of proteins includes lymphokines, interferons, colony stimulating factors and chemokines. Cytokine and chemokine research plays a significant role in achieving a deeper understanding of the immune system and its multi-faceted response to most antigens, as well as disease states such as inflammatory disease, allergic reactions, irritable bowel disease (IBD), sepsis, and cancer.

    1. During the assay validation, there was an observed increase in signal with HMGB1 when the assay buffer was used as a matrix. It was noted that HMGB1 performs optimally in a serum matrix.

  2. Does the HMGB1 antibodies used in Human Cytokine Panel 4 (HCYP4MAG-64K) detect HMGB2, HMGB3, and HMGB4?

    1. The HMGB1 capture antibody has not been tested against the other isoforms. It is a monoclonal mouse anti-Human HMGB1 with the immunogen being recombinant Human HMGB1 (Gly2-Glu215).

  3. What are the effects of Trizol in Milliplex Immunoassays known to be?

    1. Trizol has not been tested or seen in publication, so it is best not to use it in tissue homogenization. Instead, it is recommended to use a buffer that is proved compatible with immunoassays. Commonly used buffer RIPA has been shown to work well in Milliplex immunoassays.
