Other Notes

Breast cancer metastasis-suppressor 1 (UniProt: Q9HCU9, also known as BRMS1) is encoded by the BRMS1 gene (Gene ID: 25855) in human. BRMS1 is an homohexameric, predominantly nuclear protein with two nuclear localization sequences (aa 198-205 and 238-244) but may also have a cytoplasmic role. Its cytoplasmic expression has been linked to higher disease-free survival in cases of malignant melanoma. It is a member of the mSin3-HDAC transcription co-repressor complex. It is reported to suppress metastasis in multiple human and murine carcinoma cell lines by altering the expression of both coding and non-coding metastasis associated genes. It down-regulates transcription activation by NF-kappa-B by promoting the deacetylation of RelA at lysine 310 and HDAC1 binding to promoter regions. It is also shown to down-regulates expression of anti-apoptotic genes that are controlled by NF-kappa-B. HSP90 is reported to stabilize the levels of BRMS1 in cells, which may be important in maintaining the functional role of BRMS1 as a metastasis suppression.
