Interleukin-10 (IL-10), originally known as Cytokine Synthesis Inhibitory Factor (CSIF) is a protein that shares over 80% sequence homology with the Epstein-Barr Virus protein BCRFI. IL-10 down regulates the expression of Th1 cytokines, major histocompatibility complex class II antigens, and costimulatory molecules on macrophages. It also enhaces B cell survival, proliferation, and antibody production. IL-10 inhibits the synthesis of a number of cytokines, including IFN-gamma, IL-2, IL-3, TNF and GM-CSF produced by activated macrophages and by helper T-cells. IL-10 is produced by a variety of cell lines, including T-cells, macrophages, mast cells and other cell types. A polymorphism in IL10 promoter region is associated with resistance to graft-versus-host disease (GVHDS), a major complication of allogeneic bone marrow transplantation, in which mature donor T-cells that contaminate the allogeneic bone marrow recognize the tissues of the recipient as foreign, causing a severe inflammatory disease. Knockout studies suggest that IL-10 is an immunoregulator in the intestinal tract.
