General description
Apolipoprotein is a polymorphic plasma protein that exists as three isoforms: ApoE4, ApoE3, and ApoE2, which differ in the combination of cysteine and arginine residues located at position 122 and 158. The 35 kDa ApoE4 isoform contains arginine at both locations, and is produced in the liver and brain, although it is widely expressed in other tissues, such as the lung, spleen, and ovary. Like other isoforms, ApoE4 is a ligand for low density lipoprotein receptors (LDL) and plays a critical role in the transport and metabolism of lipids. In addition to its role in maintaining cardiovascular function, ApoE4 is also critically involved in neuronal plasticity, synaptogenesis, aging, and neurodegenerative disorders. Mice deficient in ApoE4 show age-dependent abnormalities in synaptic structure and impaired long term potentiation. Additionally, ApoE4 has a well-documented role in the pathology of Alzheimer’s disease.
Specificity
This antibody recognizes ApoE4 but not ApoE3 or ApoE2.
Immunogen
KLH-conjugated linear peptide corresponding to human ApoE4.
Application
Immunohistochemistry Analysis: A 1:2,000-4,000 dilution from a representative lot detected ApoE4 in human Alzheimer′s diseased brain and human cerebellum tissues.
Immunoprecipitation Analysis: A representative lot was used by an independent laboratory to detect ApoE4 in CHO cells secreting ApoE4. (Image courtesy of Dr. Egon Ogris, Medical University of Vienna)

Apolipoprotein E (ApoE) plays multiple roles in lipid transport, neuronal signaling, glucose metabolism, mitochondrial function, and inflammation in the brain. It is also associated with neurodegenerative diseases, and its influence differs depending on the isoform. In particular, the ε4 allele

White matter degradation in the frontal lobe is one of the earliest detectable changes in aging and Alzheimer disease. The ε4 allele of apolipoprotein E (APOE4) is strongly associated with such myelin pathology but the underlying cellular mechanisms remain obscure.
