General description

Amicon® Ultra 4 mL centrifugal devices with Ultracel® 3,000 Dalton molecular weight cutoff filters provide fast ultrafiltration, and reliable retention of macromolecules. For example, Cytochrome c (12.4 kDa) at 0.25 mg/ml initial concentration is retained at >95% .

The vertical design and available membrane surface area provide fast sample processing, high sample recovery (typically greater than 90% of dilute starting solution), and the capability for 80-fold concentration. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical deadstop in the filter device prevents spinning to dryness and potential sample loss.

The concentrate is collected from the filter device sample reservoir using a pipetter, while the ultrafiltrate is collected in the provided centrifuge tube. The device can be spun in a swinging bucket or fixed angle rotor. With their vertical membrane design and deadstop, Amicon® Ultra 15 mL filters deliver the highest recovery with short spin times.

Application

  1. Good day, I would like to know if these concentrators are sterile and the recovered proteins are suitable for downstream cell culture assays. Thank you!

    1. The Amicon Ultra devices are non-sterile. However, many customers rinse with 70% IPA then discard, let dry, then introduce a sample to the product.

  2. What could be the reason for no protein recovery (in the concentrate or flow through) while using the UFC800324 to purify protein? Could the protein may be absorbed in the filter?

    1. The low or no protein recovery issue may be attributed to over-concentration of the protein, leading to precipitation. The User Guide provides guidance on maximizing sample recovery: Low sample recovery in the concentrate may be due to adsorptive losses, over-concentration, or passage of sample through the membrane. Adsorptive losses depend upon solute concentration, its hydrophobic nature, temperature and time of contact with filter device surfaces, sample composition, and pH. To minimize losses, remove concentrated samples immediately after centrifugal spin. If the starting sample concentration is high, monitor the centrifugation process in order to avoid over-concentration of the sample. Over-concentration can lead to precipitation and potential sample loss. If the sample appears to be passing through the membrane, choose a lower MWCO Amicon device.

  3. What is max g force these filters can take? i.e. what sort of rpm or g force should I spin at?

    1. The UFC8003 protocol states to spin at 7,500 × g maximum for approximately 10–40 minutes. See the image below for further details.

  4. Can we centrifuge these tubes in a normal 15 mL swinging bucket centrifuge?

    1. Yes, the device can be used in centrifuges with a swinging bucket or fixed-angle rotor.
