General description

m-EnterococcusAgar, modified, is a selective dehydrated culture medium formulated for thedetection and enumeration of enterococci, particularly in water analysis. Theformulation contains peptones, yeast extract, esculin, sodium azide, nalidixicacid, cycloheximide, and agar, providing a highly selective environment thatsupports the growth of enterococci while inhibiting a wide range of Gram‑negative bacteria, fungi,and many other competing floras. Pancreatic digest ofgelatin and yeast extract acts as a source of carbon, nitrogen, minerals,vitamins, and other essential nutrients for growth. Sodium chloride maintainsisotonic conditions of the medium, besides the provision of essential ions to avariety of organisms. Sodium azide, Cycloheximide, and Nalidixic acid inhibitmany species of bacteria and fungi and thus make the medium selective. Esculinis hydrolyzed by bacterial enzymes to esculetin and dextrose. TTC is reduced byEnterococci to insoluble formazan inside the bacterial cells, which gives redcoloration to colonies

Application

m-Enterococcus Agar, modified, is a selective medium for the enumeration and detection of enterococci in water and sewage with the membrane-filter technique and in the direct application procedure in food. It is intended for the recovery and enumeration of enterococci from water samples using the membrane filtration technique and is used in conjunction with Esculin Iron Agar for confirmation and identification of typical colonies. It is suitable for monitoring drinking water, recreational water, and other environmental water samples within microbiological water quality testing schemes where enterococci serve as indicators of fecal contamination. The selectivity provided by sodium azide and nalidixic acid allows for reliable recovery of enterococci even in samples with mixed flora, supporting quantitative assessments and regulatory compliance testing in environmental and public health laboratories.

Preparation Note

Suspend 71.4 g in 1 litre distilled water. Boil to dissolve the medium completely. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 45°C and aseptically add 15 ml of sterile 1% TTC Solution Cat. No. No. 17779). Mix well and pour into sterile petri plates. Warning: Cycloheximide is very toxic. Avoid skin contact or aerosol formation and inhalation.
