Application

The addition of formamide may enhance the specificity and efficiency of amplification of PCR (polymerase chain reaction). It may be used as an ammonia equivalent for the palladium-catalyzed aminocarbonylation of 1-bromo-3-trifluoromethylbenzene to form 3-(trifluoromethyl)benzamide.
Formamide destabilizes nucleic acid duplexes and may be used, typically, at a concentration of 50%, in hybridization protocols requiring lower hybridization temperatures.

  1. What is the best media for diluting this solution?

    1. This material is miscible with water, methanol, ethanol, acetone, acetic acid, dioxane, ethylene glycol, glycerol, and phenol. This product has not been deionized, which is required for use in molecular biology applications. For use in nucleic acid hybridization and in gel loading solution, the following protocol is suggested for deionization:

      1. Mix 1 gram of a mixed bed resin (such as Sigma Prod. No. M8032) with 10 mL of formamide.
      2. Stir for approximately 1 hour.
      3. Filter two times through Whatman #1 filter.
      4. Sterile filter and store for a single use at -20 C; stable up to 6 months.

      If the deionized formamide is stored at room temperature, deionize every 2 weeks. The pH of a 0.5 M solution is approximately 5.0 before deionization and approximately 6.8 after deionization. Formamide with a yellowish appearance requires deionization.
