This product is a Saline-Tris-EDTA (STE) buffer that is ready-to-use. It has a pH specification of 7.6 to 8.0, with a lot-specific pH provided on the certificate of analysis (CofA). This buffer is prepared using Tris base, which has a pH of about 11 in water; the pH is then lowered to the 7.6 to 8.0 range by using hydrochloric acid. Since the pKa of Tris is 8.1, one could say that the buffer is 10 mM in Tris, and just a little over 5 mM in HCl.

Why do we prepare the buffer using Tris‑HCl and then adjust the pH to 8, instead of starting directly with Tris base since it is already closer to pH 8?

This product is a Molecular Biology grade. The solution contains NaCl at 100 mM, Tris HCl at 10 mM, and Na2EDTA at 1 mM. The pH range is 7.6 - 8.0. The product has been filtered using a 0.22 um pore size filter membrane and is tested for RNase, DNase, Protease and Phosphatase activity.
