Application

N-Acetyl-L-cysteine has been used to suppress oxygen-dependent hydroxyurea sensitivity in Saccharomyces cerevisiae.[1] It has also been used as an antioxidant for the treatment of C2C12 murine myoblasts.[2]

Biochem/physiol Actions

Antioxidant and mucolytic agent. Increases cellular pools of free radical scavengers. Reported to prevent apoptosis in neuronal cells but induce apoptosis in smooth muscle cells. Inhibits HIV replication. May serve as a substrate for microsomal glutathione transferase.

  1. Can this NAC be used in antimicrobial susceptibility testing?

    1. N-Acetyl-L-cysteine (NAC) is primarily known as a mucolytic agent and antioxidant, but it has also been investigated for its potential role in antimicrobial susceptibility testing. NAC can enhance the effectiveness of certain antibiotics, particularly against biofilm-forming bacteria, by disrupting biofilms and increasing the permeability of bacterial cell membranes. This property can make bacteria more susceptible to antimicrobial agents. This application has not been tested or validated internally and would need to be confirmed empirically by the end user.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

    1. This product has not been tested for in vivo or in vitro use. Stock solutions must be filter-sterilized and the endotoxin has not been determined. However, this item has been used successfully in animal models by other researchers, as listed in the Peer Reviewed Papers section of this page.   4. When you refer that NAC is soluble with heating, what temperature are we talking about? And What is the maximum temperature?

    1. Typically, NAC can be effectively solubilized in water at elevated temperatures, such as 37°C (body temperature) or higher. For optimal solubility, heating the solution to around 60-70°C can help dissolve larger quantities of NAC more easily. It's important to note that while heating can enhance solubility, care should be taken to avoid excessive temperatures that could degrade the compound.

  5. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.     1. Solubility
      - N-acetyl-L-cysteine is soluble 1 in 8 of water and 1 in 2 of ethanol.
      - It is practically insoluble in chloroform and ether.
      - The solubility of product A7250 in water at 100 mg/mL was tested with heating and resulted in a clear, colorless solution.

      Solution Stability

      Aqueous stock solutions are stable for up to 1 month at -20°C. For N-acetyl-L-cysteine, the following information is relevant:
      - Aqueous solutions of cysteine oxidize to cystine upon exposure to air at neutral or alkaline pH.
      - Oxidation is hastened by traces of heavy metals, particularly copper and iron; however, it is relatively stable in an acidic environment.
