General description

Orcein or CI Natural Red 28 is an elastic tissue stain. The fluorescence properties of Orcein show maximum emission peak at 587 nm (low concentration) and 620 nm (high concentration) upon suitable excitation and acidic pH.

Application

Orcein is highly recommended as a vital stain for elastic fibers and lamellae. Orcein is used as a nuclear stain for mounting embryos and assessing the maturation of oocytes. Other routine applications of orcein involve the staining of plant chromosomes.
