Description

General description

Safranin O is a cationic dye, which displays less significant metachromasia. It is used to detect any variations in articular disease. Safranin O binds specifically to polyanions. It detects the amount of proteoglycan present in cartilages. Safranin O is used as a marker for glycosaminoglycan depletion in osteoarthritis. Safranin is used as a counter stain in Gram staining.

Application

Safranin O has been used in the histological analysis of femurs. It has also been used to detect the presence of glycosaminoglycans in the inner body of meniscus structures.

Do you have any recommendations for the storage temperature and corresponding shelf life of Safranin O after preparation?

  As Safranin O can be prepared in a variety of solvents and at varying concentrations, it is really not possible to give precise guidelines for solution stability.
  There is a Safranin O solution that is part of a Gram Stain kit (HT90A). According to the kit Instructions For Use, this solution has 0.6% dye in 20% SD3A reagent alcohol. The expiration date for this solution is 24 months. But if the formulation varies from this exact composition or if other dyes or chemicals are also present, the 24 months expiration dating may or may not be appropriate

  Safranin O is noted for its use in visualizing and quantifying the presence of glycosaminoglycans (GAGs) within a tissue sample. However, this application is not specifically tested, and no protocol is available to recommend. It is advised to refer to academic literature for guidance.
