General description

DAB Substrate Kit is intended for use in immunohistochemistry (IHC) staining protocols. This reagent is designed to detect target antigens in formalin-fixed, paraffin-embedded tissue sections when used in conjunction with primary antibodies and appropriate secondary antibody conjugates in the IHC staining process. When in the presence of HRP polymer, DAB produces a brown precipitate that can be readily visualized using light microscopy. Kits contain: DAB chromogen and DAB buffer substrate.

Questions

1–3 of 3 Questions
  1. how to prep DAB?

    1 answer
    1. it is one drop of chromogen to 1ml of substrate buffer.

      Helpful?

  2. Is the absence of color normal when 1 drop of chromagen and 1 ml of substrate are used, given the usual light purple color?

    1 answer
    1. Ensure thorough mixing of the chromagen before combining. The color outcome is influenced by the chromogen's age. A lack of color typically suggests very recent or fresh DAB, which has not undergone oxidation.

      Helpful?

  3. What is the volume for "1 drop" of DAB chromogen?

    1 answer
    1. The volume used to prepare 1ml of DAB would be 0.006mls, rounded to 0.01mls based on calculations for the number of tests in the kit 957D-20.

      Helpful?
