Retrieves DNA fragments trapped in low-melting point agarose. The recovered DNA fragments are suitable for restriction digest, ligation and cloning.

Biochem/physiol Actions

Agarase digests low melting point agarose by attacking the unsubstituted carbohydrate moieties of neogarobiose agarose (3,6-anhydro-α-L-galactopyranosyl-1,3-D-galactose) and releasing neogaro-oligosaccharides. The mixture of agarase and released carbohydrates does not inhibit restriction endonuclease activity or prevent DNA fragment ligation and transformation.


Tested for suitability in isolating λ DNA Hind III fragments from low-melting agarose gels.

Unit Definition

One unit will digest 100 μl of molten 1% low-melting agarose.
