General description

Interleukin-10 (IL-10) is a pleotropic cytokine with anti-inflammatory and immunosuppressive functions. It is the key cytokine that regulates the host inflammatory responses to bacterial, fungal and viral pathogens. The production of IL-10 is mediated by signalling of IL-6, IL-12 and TGF-β. IL-10 is secreted by a variety of cells including T cells, B cells, dendritic cells, eosinophils, natural killer cells and mast cells. The effects of IL-10 are mediated by binding to the IL-10R, consisting of α and β subunits. The signalling through IL-10R activates Jak1, Tyk2 protein kinases and JAK/STAT3 pathways. IL-10 is involved in suppression of innate immunity to kill pathogens and inhibits the release of reactive oxygen and nitrogen species from activated macrophages. Regulation of IL-10 production is important in infections, inflammatory responses in autoimmune encephalomyelitis, endotoxic shock and chronic enterocolitis
Anti-interleukin-10 antibody is specific for mouse IL-10. The antibody shows less than 5% cross reactivity to recombinant human IL-10.

Specificity

The antibody neutralizes the biological activity of recombinant mouse IL-10.

Immunogen

recombinant mouse IL-10 expressed in E. coli.

Application

Anti-interleukin-10 antibody may be used for immunoblotting at a working concentration of 1-2 μg/ml. The antibody is suitable for neutralization reactions (ND50 is 2-4 μg/ml).

Physical form

Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline with 5% trehalose.

Preparation Note

Purified using protein G.

Understanding the mechanisms that drive transition from acute to chronic pain is essential to identify new therapeutic targets. The importance of endogenous resolution pathways acting as a "brake" to prevent development of chronic pain has been largely ignored. We examined...

In the present study, we define the relation between TGF-beta and IL-10 in the regulation of the Th1-mediated inflammation occurring in trinitrobenzene sulfonic acid (TNBS)-colitis. In initial studies, we showed that the feeding of trinitrophenol-haptenated colonic protein to SJL/J mice...

IL-10 plays an essential part in controlling inflammation and instructing adaptive immune responses. Consequently, dysregulation of IL-10 is linked with susceptibility to numerous infectious and autoimmune diseases in mouse models and in humans. It has become increasingly clear that appropriate...

The generation of an effective immune response against an infection while also limiting tissue damage requires a delicate balance between pro- and anti-inflammatory responses. Interleukin-10 (IL-10) has potent immunosuppressive effects and is essential for regulation of immune responses. However, the...
