Application

Corticosterone has been used:
  • in chronic treatment to investigate its effect on somatostatin (SST) neurons in mice[1]
  • as a stress inducing hormone in pregnant mice[2]
  • to test its effect on depression-related behaviors in mice[3]

Biochem/physiol Actions

Corticosterone is a glucocorticoid secreted by the adrenal cortex that activates both mineralocorticoid and glucocorticoid receptors.
Corticosterone is synthesized from 11-deoxycortisone by the action of enzyme 11β-hydroxylase majorly in adrenal glands.[4] However, it may also be synthesized in non-adrenal tissues including leydig cells of testes[4] and in murine thymus.[5] Corticosterone is released post activation of hypothalamus-pituitary-adrenal (HPA) axis. It is a key player regulating metabolism, immune function and response to stress.[6] It is produced by HPA activation in response to stress in myeloid cells.[7] Elevated levels of corticosterone is observed in Huntington′s disease (HD).[8]

  1. I tend to treat pc12 cell line with CORT C2505, and the question is how can I reconstitute the powder to be non-toxic to the cells? Because the chloroform or ethanol are toxic to the cells

    1. Corticosterone is also soluble in DMSO at 69 mg/mL ( (199.15 mM). It is best to use a fresh stock bottle of DMSO that is deemed free of any moisture. Any contaminating moisture may accelerate the degradation of the compound in question or may render it insoluble. Please make sure the final concentration of DMSO in cell culture should not exceed more than 0.1%.

  2. Which reagents should be used to dissolve product C2505, and what are the recommended storage conditions after it has been dissolved?

    1. C2505 is soluble in Chloroform at 50 mg/mL, and should be colorless to faintly yellow in appearance.

      Although we cannot provide any verified recommendations for temperature storage of solution, other literature sources suggest that the solution can remain at room temperature if the solution stays clear for 3 days. These sources suggest that it is soluble in ethanol at 50 mg/ml, and that following reconstitution, aliquot and freeze (-20°C), for stock solutions to be stable for up to 3 months (at -20°C). There are additional sources that recommend dissolving in acetone, methanol, ethanol (10 mM), chloroform, and DMSO (100 mM). It is insoluble in water.
