Application

Cholesterol-Water Soluble has been used:
  • in free cholesterol (FC) overloading of rat smooth muscle cells (SMCs)
  • in cytotoxicity assay
  • for the incubation of CD4+ T cells with Helicobacter pylori[1]
  • to study the membrane cholesterol level in SH-SY5Y human neuroblastoma cells
  • to study the effects of cholesterol on cell viability and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction in primary cultured rat hippocampal neurons
  • as a supplement for growth of bacterial culture
  • to examine the interaction of S protein with angiotensin-converting enzyme 2 (ACE2)

  1. How stable is this product when dissolved in water to 300 mg/mL? Can it be frozen for long-term storage?

    1. Solutions should be stored in single use aliquots at -20ºC. Frozen aliquots should be stable for at least 6 months.

  2. What is the molecular weigth of this specific product? Can you share and add g/mol vale please?

    1. The molecular weight of cholesterol used for this product is 386.65 g/mol. For the calculation of molarity based on cholesterol content, approximately 0.4 mg of the 10 mg vial content is cholesterol. The lot-specific Certificate of Analysis should be checked to find the lot-specific cholesterol content for the received product.

  3. Hi, When you say it has biphasic solubility, I understand what you mean, but in that, when you refer to the mg, do you mean the mg of cholesterol WITHIN the compound methyl-B-cyclodextrin, or the mg of the entire compound?

    1. The reference to mg pertains to the amount of solid powder used, which is 300 mg of the product per mL of water. It is important to allow the solution to sit undisturbed for one hour after dilution to achieve complete solubilization of the product.

  4. Is there a minimum or maximum solubility limit for this Cholesterol-Cyclodextrin product?

    1. Due to the nature of cyclodextrin, the cholesterol-cyclodextrin complex exhibits biphasic solubility. To ensure optimal solubility at low concentrations, it is suggested that stock solutions be prepared at a minimum of 50 mg/mL   Note that the optimum suggested stock solution concentration is 100 – 300 mg/mL.   This material has not been tested for solubility at concentrations greater than 300 mg/mL.

  8. Hay algo que no entiendo respecto a este producto. Se asegura que el contenido es de 40 mg de colesterol por g de producto, sin embargo el envase es de 30 mg, eso quiere decir que la concentración de colesterol en el envase es de 1,2 mg? no queda claro

    1. The current specification for this product is that it contains greater than or equal to 40 mg of cholesterol per gram. Each vial contains 30 mg of cholesterol. Therefore, the total mass of the product will always be less than 750 mg.

    1. The bulk of this material is Methyl-?-cyclodextrin. This component enhances the solubility of Cholesterol in water. There are approximately 40 mg of Cholesterol per gram of product, with the remainder being the cyclodextrin agent. Cyclodextrins are not cell permeable and inert in culture.   10. CAN CHOLESTEROL POWDER BE DISSOLVED IN Medium of cells (High glucose, 2%fbs, 1% p/s) ?

    1. The product is tested for solubility in water at 300 mg of the powder per 1 mL. Reconstituting in medium or salt solutions may cause a precipitate and is not recommended. Stock solutions in water may then be filter sterilized and added directly to the culture medium. Note that this product is sold on the basis of the cholesterol content only. The 30 mg package size may contain nearly 1 gram of powder. The specific amount of cholesterol per gram is reported on the lot-specific Certificate of Analysis.     Not Water Soluble

    I attempted to use this product but it is not water soluble as advertised. I followed the protocol that is given on the website, mixing in water and letting the mixture sit undisturbed for 1 hour. Afterwards, there were still many undissolved crystals and the mixture could not be filtered without severely diluting our mixture. We also tried letting the mixture sit in a 37 degree water bath for 30 minutes, followed by letting it sit overnight in a 4 degree fridge, and the cholesterol was still not dissolved.

      This issue may be due to the solution concentration being too high (>300mg/ml) or too low. Due to the nature of cyclodextrin, the cholesterol-cyclodextrin complex exhibits biphasic solubility. To ensure optimal solubility at low concentrations, it is suggested that stock solutions >50 mg/mL (and preferably >100 mg/mL) be prepared, and then diluted to the desired concentration. Also, addition of un-complexed cyclodextrin (product C0926) aides solubility at low concentrations.

    Not soluble in water

    Even 2mg/ml is not soluble in deionized water. I see crystals
