Cell Line Origin

Human neuroblastoma

Cell Line Description

Spontaneously transformed cells from a female patient with neuroblastoma. The tumour was derived from a bone marrow metastasis and is tumourigenic in nude mice. Used in the study of MIBG uptake in the diagnosis and treatment of neuroblastoma. MIBG is a derivative of the antihypertensive agent guanethidine and is used as a radiopharmaceutical. SK-N-DZ is characterised by low saturable kinetics and predominant linear pattern of uptake of MIBG, with a poor ability to maintain high intracellular levels of the drug.

Application

Tumour studies. MRD-1 resistance studies. Extremely sensitive to cis-retinoic acid.

Culture Medium

DMEM or EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:10 i.e. seeding at 1-3x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37°C.
