General description

α1-Acid Glycoprotein (AGP, orosomucoid) from bovine plasma belongs to ‘lipocalins′ family which binds and transports small hydrophobic molecules. AGP is further classified into the subfamily ‘immunocalins′ which immunomodulates the inflammatory reactions.[1] AGP is primarily produced in the liver, in few extra hepatic tissues and is present in neutrophil granules. Healthy cattle possesses 0.3 mg of AGP/ ml of concentrated serum.[2]

α1-Acid glycoprotein (AGP, orosomucoid) is the major plasma glycoprotein with a molecular weight of 41-43 kDa. It is responsible for possessing the complex type of glycans.^[3]

Application

α1-Acid Glycoprotein (AGP) from bovine plasma has been used:

in mass spectrometry analysis to confirm the presence of Neu5Ac and Neu5Gc distribution in human and bovine AGP respectively[4]
for pretreating the microvessel before perfusing the MDA-MB-231 cells for tumor adhesion studies[5]
in the composition of tear model[6]
as a reference in sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE)[7]

α1-Acid glycoprotein was used to study nonviral gene delivery to the lung with copolymer-protected and transferrin-modified polyethylenimine^[8]

Biochem/physiol Actions

α1-Acid Glycoprotein from bovine plasma is an acute phase protein. It has anti-inflammatory potential and its concentration increases several folds during injury. AGP increases the chemotactic activity and life span of monocytes and inhibits neutrophil degranulation.[2] Serum AGP levels differentiates sepsis from systemic inflammatory response syndrome (SIRS).[9]
