DEAD (Asp-Glu-Ala-Asp) box helicase 3 (DDX3X) is an ATP-dependent RNA helicase. It has multiple conserved domains and has roles in both the nucleus and cytoplasm.

DEAD (Asp-Glu-Ala-Asp) box helicase 3 (DDX3X) has RNA-unwinding and adenosine triphosphatase (ATPase) activities. The other functions include nuclear mRNA export, cell growth control and cancer progression. The ATPase activity of the helicase is stimulated by various ribo- and deoxynucleic acids. Studies with different nucleoside triphosphate analogs showed that the human DDX3 ATPase has high catalytic efficiency and rather relaxed substrate specificity, both in terms of base selection and sugar selection. It has an ability to recognize the L-stereoisomers of both 3′ deoxy- and 2′, 3′ dideoxy-ribose, showing it has low stereo selectivity.
