General description

ITS+3 liquid media supplement is a mixture of insulin, human transferrin (partially iron-saturated), sodium selenite, linoleic acid, and bovine serum albumin (BSA). It is suitable for serum-free cell culture applications.

Application

ITS+3 Liquid Media Supplement (100×) has been used as a media supplement:
  • to support the growth of human adipose stem cells (ASC)
  • to culture porcine intervertebral disc (IVD) cells and tissues
  • to culture cartilage endplate (CEP) cells

Biochem/physiol Actions

  • Insulin: a polypeptide hormone that promotes glucose and amino acid uptake and may produce a mitogenic effect.
  • Transferrin: an iron-transport protein. Iron is an essential trace element but can be toxic in its free form. To nourish cells in culture, iron is supplied bound to transferrin in serum.
  • Selenium: an essential trace element present in serum.

Components

Contains 1.0 mg/ml bovine insulin, 0.55 mg/ml human transferrin (substantially iron-free), 0.5 μg/ml sodium selenite, 470 μg/ml linoleic acid, 470 μg/ml oleic acid and 50 mg/ml bovine serum albumin at the 100x concentration. Contains 2 moles each of linoleic acid and oleic acid per mole of albumin.

Preparation Note

Prepared in Earle′s Balanced Salt Solution (EBSS) without phenol red.
