Application

Lipase from porcine pancreas has been used:
  • for the analysis of fatty acid at the 2-position of triglycerides.[1]
  • to determine the indigestible fraction of plant source that is resistant to the action of digestive enzymes.[2]
  • as a component of dissolution media to study the dissolution profiles of theophylline.[3]
Lipases are used industrially for the resolution of chiral compounds and the transesterification production of biodiesel.

Biochem/physiol Actions

Lipases catalyze the hydrolysis of esters in aqueous solutions and the synthesis of esters in non-aqueous solutions. They also produce hydroxyl and carboxylic groups through the hydrolysis of ester linkages in poly(ethylene terephthalate).

Lipases catalyze the hydrolysis of triacylglycerols into glycerol and free fatty acids.

Quality

Crude (Steapsin). Contains amylase and protease activity.

Unit Definition

One unit will hydrolyze 1.0 microequivalent of fatty acid from triacetin in 1 hr at pH 7.4 at 37°C. (pH 7.7 is used with olive oil as substrate)

    1. This product contains 50% (v/v) olive oil (catalog number O1500).
      1% sodium azide is added as a preservative and an emulsifier. The exact composition and method of
      composition is considered proprietary and therefore will not be disclosed.

  3. I want to use this product (L3126) for preparing a solution to conduct in vitro degradation experiments on a particular material. Could you please provide detailed instructions on how to prepare the solution, including any recommendation?

    1. Unfortunately, there is no application specific instructions available for this product. The material may be reconstituted in 100 mM potassium phosphate buffer, pH 8.0, containing 1 mg/ml BSA. This product is from porcine pancreas. Another product option, L3419 Lipase from Thermus thermophilus, is reconstituted in 50 mM Tris-HCl, pH 8.0. This material states that solutions are stable for up to 1 month at 2 - 8°C. Please see the link below to review the datasheet for this item:

    1. A microequivalent is equivalent to 1 mmol/L. In this case, one unit of Lipase will hydrolyze 1.0 microequivalent (1 mmol/L) of fatty acid from olive oil in 1 hour at pH 7.7 at 37°C.
