Application

Ultra-low Range Molecular Weight Marker (M.W. 1,060-26,600) has been used as a protein marker in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and western blot.[1]

Biochem/physiol Actions

The ultralow range marker is designed for use in the Tris-Tricine SDS-PAGE method when stained with Brilliant blue G (non-methanol based). Glutaraldehyde fixing is recommended with the ultralow marker in order to keep the two smallest proteins fixed in the gel. The ultralow range marker is supplied as a liquid, which must be diluted (1:20) with the supplied sample buffer prior to loading.

Physical form

Solution in 0.1 M Tris-HCl, pH approx. 8.5, with 4 mM EDTA, 3 mM sodium azide and 40% glycerol

Questions

  1. Can I use it for Coomassie blue stain SDS Page (16.5%)? Just to confirm: Is it for 1kDa to 26kDa range of proteins?

    1 answer
    1. Technically, the Ultra Low Range Molecular Weight Marker (M.W. 1,060–26,600), Sigma Catalog Number M3546, can be used with Coomassie Blue staining on a 16.5% SDS-PAGE gel. Since we do not have compatibility data with Coomassie blue staining, the customers will need to validate this on their behalf.

Each vial of marker contains 200 ml of a mixture of six peptides and proteins (see Table 1) in 100 mM
      Tris-HCl, pH 8.5, 4 mM EDTA, 3 mM sodium azide, and 40% glycerol. The 2x sample buffer is a solution of 100 mM Tris-HCl, pH 6.8, 1% SDS, 4% 2-mercaptoethanol, 0.02% Brilliant Blue G, and 24% glycerol.
