General description

The mouse Anti-c-MYC monoclonal antibody recognizes an epitope located within a sequence (EQKLISEEDL) of the product of the human oncogene c-myc.
c-Myc (v-myc avian myelocytomatosis viral oncogene homolog) is a transcription factor, which is coded by MYC gene. It was first described as a homolog of an avian retroviral oncogene. It belongs to the family of transcription factors which are characterized by basic region helix-loop-helix/leucine zipper.[1] Along with N-Myc and L-Myc, it forms the MYC proto-oncogene family.[2] It forms a heterodimer with MAX, and this heterodimer binds to the E-boxes/CACGTG sequence motif on DNA.[1] This gene is localized to human chromosome 8q24.[3]

Application

Monoclonal Anti-c-Myc antibody produced in mouse is suitable for immunoprecipitation,[4] immunoblotting,[5][6][7] ELISA, immunofluorescence,[8][9] microarray and electron microscopy. It has also been used in ChIP (Chromatin Immunoprecipitation).[10][11]

Biochem/physiol Actions

v-myc avian myelocytomatosis viral oncogene homolog belongs to MYC family. When overexpressed these proteins can cause DNA double-stand breaks (DSBs) and genome instability.[12] This transcription factor MYC helps in regulating long noncoding RNAs (lncRNAs) which has been implicated in cancer cell proliferation and tumorigenesis.[13] Its overexpression is also associated with various cancers, and in particular B cell lymphomas. Increased expression of c-MYC has a negative effect on the ability of B cell lymphomas to functionally present Ags/peptides to CD4+ T cells. This protein exerts suppressive effects at several critical checkpoints in Ag presentation.[1] It is associated with Burkitt lymphoma and a group of diffuse large B-cell lymphoma (DLBCL).[14]
