General description

The MOPC-21 tumor line that produces mouse IgG1, κ is a mineral oil induced plasmacytoma originated and carried intraperitoneally in BALB/c mice. Immunoglobulins G are made up of two identical light chains and two identical heavy chains, that are covalently connected by interchain disulphide bonds.[1] Mouse IgG is further divided into five classes- IgG1, IgG2a, IgG2b, IgG2c and IgG3.[2]

Application

IgG1, Kappa from murine myeloma has been used:
  • in stimulation assay and enzyme-linked immunosorbent assay (ELISA)[3]
  • as isotype controls in islet hormone production[4]
  • in flow cytometric analysis[5], in cell migration assay[6]
  • as control antibody in evaluation of methylation[7]

Biochem/physiol Actions

IgG antibody subtype is the most abundant of serum immunoglobulins of the immune system. It is secreted by B cells and is found in blood and extracellular fluids. It provides protection from infections caused by bacteria, fungi and viruses. Maternal IgG is transferred to fetus through the placenta that is vital for immune defense of the neonate against infections.[8]
Immunoglobulin G (IgG) participates in hypersensitivity type II and type III reactions.[9] IgG helps in opsonization, complement fixation and antibody dependent cell mediated cytotoxicity.[2]

Preparation Note

Reconstitute with 1 mL deionized water.

Questions

  1. Good morning, I have a question about product specification. Looks like 1vial contains about 5mg and the protein, correct? What does the 15mg/ml protein by Biuret info mean? Because if the reconstitution is done in 1mL, the highest conc. is 5mg/mL, no? Ty

    1 answer
    1. This product is lyophilized from mouse ascites fluid. The total protein content is verified prior to lyophilization. The 15 mg/mL value is the pre-lyophilization total protein content. The Myeloma Protein-Densitometry is performed after lyophilization when reconstituted to 1ml in water. The 5 mg value is specific to the Myeloma Protein (IgG1 kappa) only.
