General description
Cholesterol is predominantly produced in the liver.[4] Abnormal cholesterol levels are implicated in coronary heart disease.[5] Cholesterol is associated with breast cancer progression or inhibition. Tumor cells are capable of synthesizing their own cholesterol, which aids in cell proliferation.[4] Increased cholesterol level is observed in cancers, especially in prostate cancer.[6] Elevated serum cholesterol is observed in obesity.[5]

How is shipping temperature determined? And how is it related to the product storage temperature?

Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. Hi, Can the cholesterol quantification assay (MAK043) measure the cholesterol content in lipid nanoparticles? Does it break down the particles and measure total cholesterol or does it measure only unreacted cholesterol?

The Cholesterol Quantitation Kit can also determine the concentration of free cholesterol, cholesteryl esters, or both (total) present in a sample. The assay is enzyme-based and needs the cholesterol or cholesterol esters in solution rather than in a membrane type structure. The kit has not been tested directly for use with lipid nanoparticles.

Can you use this kit on frozen animal cell pellets?

It is recommended to use fresh samples for this kit. However, although not ideal, flash-frozen tissue or cells stored at -80C can be used. It is important to remember that results may vary between fresh and frozen samples. If possible, fresh and frozen samples should be run parallel to compare results.

Can this kit be used with bacterial supernatant (cultivation medium without cells) and how prepare these samples? Is necessary organic extraction?

This product has not been tested for use in bacterial culture supernatent, however there is no reason to suspect that this kit would not be suitable. Solid tissues and cell lysates are extracted using chloroform:isopropanol:Igepal preparation. Samples such as serum, plasma, or other biological fluids do not require organic extraction. The minimum concentration for detection has not been determined, although the first data point in the standard curve is 1 ug/well for the colorimetric assay and 0.1 ug/well for the fluorometric assay. What's the limit of detection (LOD) and detection range for cholesterol quantitation kit?

The absolute limit of detection of this kit is not determined. However the kit is able to quantify at least 1 ug/well (colorimetric) or 0.1 ug/well (fluorimetric).

Can these kits be used with serum samples from mouse?

Yes, this kit can be used on mouse serum samples. We generally recommend that fresh serum samples be used. Frozen samples can probably be used. However, the successful use of frozen samples depends on the efficiency of freezing them and the amount of time they have been frozen. If previously frozen serum samples are used, you may want to check the performance of the kit with fresh vs. frozen samples.
