Gene Information

General description

Pyruvate is a central molecule in metabolism that is used and synthesized through multiple metabolic pathways including glycolysis and the transamination of alanine. Pyruvate may be further oxidized in the citric acid cycle, converted to carbohydrates during gluconeogenesis, or reduced to lactate. High levels of pyruvate are associated with liver disease and genetic disorders. Pyruvate has also been used to stimulate metabolism leading to loss of body weight.

Suitability

Suitable for measuring pyruvate concentration in various biological samples such as blood, tissues, and culture cells.

Principle

Pyruvate concentration is determined by a coupled enzyme assay, which results in a colorimetric (570 nm)/fluorometric (λex = 535/λem = 587 nm) product, proportional to the pyruvate present.

Microorganisms have evolved to produce specific end products for many reasons, including maintaining redox balance between NAD+ and NADH. The yeast Saccharomyces cerevisiae, for example, produces ethanol as a primary end product from glucose for the regeneration of NAD+. Engineered

Questions

  1. Can I warm my pyruvate Assay buffer to 37°C to dissolve the precipitated flakes at the bottom, or will that disrupt the assay?

    1 answer
    1. All components should be thawed at room temperature. If the components are fully thawed at room temperature and there is still precipitate in the Assay buffer, follow this step: It is advised that the buffer can be kept at 37°C for a short time to see if it helps. This brief exposure to 37°C should not negatively affect the assay; in fact, many researchers keep the buffer bottle at this temperature in a water bath while thawing it.
