This kit is compatible with cell samples, and the supplied reagent component can be used to lyse the cells. The following protocol outlines sample preparation techniques for measuring intracellular heme:
1. Pellet and wash the cells.
2. Homogenize the cells with the reagent containing NaOH and detergent, which will effectively lyse the cells.
3. Incubate for 5 minutes to allow color development, then centrifuge to pellet any debris.
4. Transfer the supernatant to a titer plate or cuvette and measure the absorbance at 400 nm.
