General description

Hemoglobin (Hb) is an iron-containing metalloprotein that serves as the primary means of oxygen transport in vertebrates. Hemoglobin is primarily found in red blood cells[1] where it makes up to 97% of the cell′s dry content. Hemoglobin can also be found in other tissues where it serves as an antioxidant. Alterations in blood hemoglobin levels occurs in many diseases such as anemia and polycytheima.[2][3]

Hemoglobin Assay Kit has been used to measure the concentration of hemoglobin.[4][5]
Suitable for the detection of hemoglobinin a variety of samples such as blood, serum, plasma, and urine. This kit can be used to study the effects of various compounds on hemoglobin metabolism and levels.

Biochem/physiol Actions

This assay is based on the improved Triton[NP1]/NaOH method in which hemoglobin is converted to a colorimetric product measured at 400 nm. This assay has a linear detection range between 0.9-200 mg/dL in the 96 well plate assay.

  1. I am doing erythroblast differentiation experiment, so that i am looking for hemoglobin content assay kit. I wonder this kit can be used for cell sample?

    1. This kit is compatible with cell samples, and the supplied reagent component can be used to lyse the cells. The following protocol outlines sample preparation techniques for measuring intracellular heme:

      1. Pellet and wash the cells.
      2. Homogenize the cells with the reagent containing NaOH and detergent, which will effectively lyse the cells.
      3. Incubate for 5 minutes to allow color development, then centrifuge to pellet any debris.
      4. Transfer the supernatant to a titer plate or cuvette and measure the absorbance at 400 nm.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. What is the concentration of the calibrator that comes with the kit? Can this assay be used with cuvettes and if so is there a protocol for reagent volumes and concentrations?

    1. The calibrator, when diluted for use, has a concentration of 100 mg/dL (100 mg / 100 ml). The kit's Technical Bulletin includes instructions for using the kit with cuvettes as well as with a 96-well plate.

  4. Is this a hemolysis kit? Certain capmounds are used to detect compatibility with human blood; is this used to detect the amount of free hemoglobin? Can this be used to find whether our compound has cytotoxicity towards RBC?

    1. The assay kit can be used to detect free Hemoglobin. It is not suitable for use in the detection of substances that are cytotoxic to red blood cells.

  5. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   6. Is there stability information available for the Reagent (MAK115A) and Calibrator (MAK115B) included in the hemoglobin assay kit, especially after they are opened?

    1. The recommended storage for both components is at 2-8 degrees Celsius when not in use. Cycling the components between refrigerated and room temperature may decrease shelf life, but there is no predicted rate of degradation. For prolonged shelf life with frequent use, aliquoting the reagents into smaller tubes for individual use is advised.

  7. What specific items are included in this kit? Is there a standardized hemoglobin assay?

    1. There is a hemoglobin calibrator included with the kit, and the concentration of the sample is determined relative to the known concentration of the calibrator.
