General description

Peroxide (e.g. hydrogen peroxide H2O2), a reactive oxygen species produced through the metabolism of molecular oxygen, serves as both an intracellular signaling messenger and a source of oxidative stress. Hydrogen peroxide is generated in cells via multiple mechanisms such as the NOX-mediated ROS production by neutrophils and macrophages (respiratory burst) or by the dismutase of superoxide anions produced as a result of electron leak during mitochondrial respiration. Abnormal hydrogen peroxide production contributes to oxidative cell damage and the progression of diseases such as asthma, atherosclerosis, osteoporosis, and neurodegeneration.

Application

Suitable for the quantitative determination of peroxide concentration without any pretreatment in a variety of biological samples, such as serum, citrate-plasma, urine, cell lysates, and culture medium.
Peroxide Assay Kit has been used to measure hydrogen peroxide level in rat tissue homogenates.

Biochem/physiol Actions

The Peroxide Assay Kit provides a simple and high-throughput adaptable assay for quantitative determination of peroxide concentration without any pretreatment in a variety of biological samples, such as serum, citrate-plasma, urine, cell lysates, and culture medium. This assay utilizes the chromogenic Fe3+ −xylenol orange reaction, in which a purple complex is formed when Fe2+ is oxidized to Fe3+ by peroxides present in the sample, generating a colorimetric (585 nm) result, proportional to the level of peroxide present. The optimized formulation reduces interference by substances in the raw samples. A concentration of 1 μM H2O2 equals 34 ng/mL or 34 ppb. The kit has a detection range of 0.2–30 μM (7–1,020 ng/mL) H2O2 in a 96 well format.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   4. Is 1% TWEEN in the sample compatible with the MAK311 peroxide assay kit?

    1. The tween in samples should remain below 0.25%. If the sample contains 1% tween it is recommend diluting 4x to 0.25%, assuming the sample has enough peroxide to still be in the detectable range. The kit has a detection range of 0.2–30 μM (7–1,020 ng/mL) H2O2 in a 96 well format.

  5. Is it possible to use the MAK311-1KT Peroxide Assay Kit to measure peroxide in lysed cells that are grown in Minimum Essential Medium Eagle containing Phenol Red at a concentration of 0.011 g/liter?

    1. The reagents used with the MAK311-1KT Peroxide Assay Kit create an acidic environment in the well plate, and Phenol Red should not have measurable absorbance at the kit's wavelength of 585 nm.
      As an extra precaution, it is recommended to dilute the H2O2 standard in the same media that is used on the cells to account for any background or chemical interactions from the media. Furthermore, it is best for samples to contain less than 1 mM pyruvate.
