Application

The Caspase-3 Assay Kit may be used for:
  • Apoptosis Inducer and Inhibitor Screening
  • Cancer Research
  • Neurological Research
  • Drug Discovery

The kit is suitable for the quantitative determination of the apoptosis target caspase-3 activity and high throughput screening for apoptosis inducers and inhibitors in cell and tissue lysate.

Biochem/physiol Actions

The Caspase-3 Assay Kit provides a convenient means to measure caspase-3 activity in biological samples. In the assay, a specific substrate (N-Ac-DEVD-AFC) is cleaved by active caspase-3, forming a highly fluorescent product. The fluorescence intensity (λEx = 400 nm/λEm = 490 nm) is proportional to the caspase-3 activity.

Features and Benefits

  • Efficient Procedure: Simplify your workflow with our hassle-free process, requiring only the addition of a single working reagent directly to your cell or lysate samples, saving valuable time and effort.
  • Compatibility with High-Throughput Systems: Seamlessly integrate our kit into high-throughput handling systems, ensuring smooth operations and accurate results, enhancing efficiency in your laboratory workflow.

General description

CPP32 is a gene encoding a member of the caspase family, crucial for both the initiation and execution phases of apoptosis, a fundamental process in various physiological and pathological events, ranging from normal fetal development to diseases such as cancer, organ failure, and neurodegenerative diseases. Human caspases are categorized into three functional groups: cytokine activation, apoptosis initiation, and apoptosis execution. Among these, Caspase 3, also known as CPP32, Apopain, Yama, and PARP cleavage protease, is extensively studied and regulates numerous cellular events. Its activity is modulated by various factors, including APAF1, CFLAR/FLIP, NOL3/ARC, and members of the inhibitor of apoptosis (IAP) family. Caspase 3 undergoes self-proteolysis and/or cleavage by other proteases, resulting in its activation. It subsequently cleaves and activates other caspases and targets in cells. Variations in Caspase 3 levels are observed in cells with different lifespans and in diseases like Alzheimer′s. Caspase-3 serves as a vital biomarker in apoptosis assessment and aids in understanding its underlying mechanisms.

Questions

  1. Is Caspase-3 Assay MAK457 compatible with flow cytometry for analysis? If yes, could you provide the mechanism or protocol for its use in flow cytometry? Thank you.

    1 answer
    1. The intended use for this kit is to assess Caspase-3 activity in a 96 well plate using a fluorometer. No specific protocol has been written for using this kit using a flow cytometer. Please contact your local TS team for specific product recommendations as it is likely there is an alternative caspase activity assay kit that has been optimized for this technique.
