In the assay, a specific substrate (N-Ac-DEVD-AFC) is cleaved by active caspase-3, forming a highly fluorescent product. The fluorescence intensity (λEx = 400 nm/λEm = 490 nm) is proportional to the caspase-3 activity.

General description
CPP32 is a gene encoding a member of the caspase family, crucial for both the initiation and execution phases of apoptosis, a fundamental process in various physiological and pathological events, ranging from normal fetal development to diseases such as cancer, organ failure, and neurodegenerative diseases. Human caspases are categorized into three functional groups: cytokine activation, apoptosis initiation, and apoptosis execution. Among these, Caspase 3, also known as CPP32, Apopain, Yama, and PARP cleavage protease, is extensively studied and regulates numerous cellular events. Its activity is modulated by various factors, including APAF1, CFLAR/FLIP, NOL3/ARC, and members of the inhibitor of apoptosis (IAP) family. Caspase 3 undergoes self-proteolysis and/or cleavage by other proteases, resulting in its activation. It subsequently cleaves and activates other caspases and targets in cells. Variations in Caspase 3 levels are observed in cells with different lifespans and in diseases like Alzheimer′s. Caspase-3 serves as a vital biomarker in apoptosis assessment and aids in understanding its underlying mechanisms.
